Indications SINTOCEF ® Cefonicid

SINTOCEF ® is indicated in the treatment of bacterial infections sustained by microorganisms sensitive to cephalosporins.

SINTOCEF ® has also been shown to be effective in pre, intra and post-operative antibiotic prophylaxis.

Mechanism of action SINTOCEF ® Cefonicid

SINTOCEF ®, an antibiotic used in the treatment of difficult infections generally supported by penicillin-resistant Gram negative bacteria, is a medicine based on Cefonicid, a semi-synthetic active ingredient belonging to the category of beta-lactam antibiotics and more specifically of second-generation cephalosporins.

In particular with these latter shares both the mechanism of action, characterized by the inhibition of the transpeptidation reaction, then by the formation of cross-linking between peptidoglycan molecules useful to stabilize the bacterial wall, both the broad spectrum of action and the natural resistance to the inhibitory action of penicillinases and bacterial beta-lactamases.

Taken intramuscularly or intravenously, Cefonoicid is distributed among the various tissues and biological fluids, with the exception of the cerebrospinal fluid, carrying out its own antibiotic action for about 24 hours after intake.

After a half-life of about 4-5 hours, it is mostly eliminated via the kidneys.

Important case report that reports the appearance of severe anaphylaxis following administration of cefonicid. Above all for these reasons it would be advisable to carry out a thorough medical history, which assesses the presence of conditions incompatible with the therapy, before taking antibiotics.

Method of use and dosage

SINTOCEF ®

Powder and solvent for solution for injection of 1 g of Cefonicid.

The particular pharmacokinetic properties of cefonocid, able to maintain the therapeutic effect for about 24 hours after its intake, allow the daily administration of SINTOCEF ® (1 gram) both intramuscularly and intravenously.

Only in very exceptional cases it may be necessary to further increase the doses, while a further adjustment could be required in elderly patients or those suffering from renal insufficiency.

In any case the administration of SINTOCEF ® should take place under strict medical supervision.

Warnings SINTOCEF ® Cefonicid

Careful medical entrance and periodic monitoring of renal function would be necessary before and during treatment with SINTOCEF ®.

Particular caution is instead indispensable in elderly and nephropathic patients, normally more susceptible to the side effects of antibiotic therapy.

Should the patient develop any type of side effect or unexpected adverse reaction, he should urgently consult his doctor with whom to evaluate the possibility of suspending treatment.

Antibiotic therapy should be prolonged for at least 48 hours after the clinical symptoms have disappeared, to avoid possible relapses.

PREGNANCY AND BREASTFEEDING

The use of SINTOCEF ® during pregnancy and in the subsequent period of breastfeeding is generally contraindicated, although it could exceptionally be tolerated in cases of real need and always under the supervision of your doctor.

Interactions

Although the pharmacological interactions worthy of clinical relevance are almost entirely absent, the patient subjected to antibiotic therapy with SINTOCEF ® should pay particular attention to the contextual assumption of potentially nephrotoxic drugs, capable of further compromising the excunctory capacities of this organ.

Contraindications SINTOCEF ® Cefonicid

The use of SINTOCEF ® is contraindicated in patients hypersensitive to penicillins and cephalosporins or to their excipients as well as to lidocaine (for intramuscular administration).

Undesirable effects - Side effects

Although in rare cases different clinical trials and careful post-marketing monitoring have shown that the intake of cephalosporins, and therefore also of cefonicid, can determine the appearance of:

  • Gastrointestinal reactions such as nausea, vomiting and diarrhea;
  • Renal disorders such as tubular and glomerular damage accompanied by urinary abnormalities and proteinuria;
  • Haematological reactions such as haemolytic anemia, neutropenia, leukopenia and myeloinhibition;
  • Liver damage observable with hypertransaminasemia and hyperbilirubinemia;
  • Lethargy, spasms, convulsions and various neurological symptoms;
  • Even severe allergic reactions such as angioedema, pruritus, urticaria, bronchospasm, hypotension and anaphylactic shock.
