Indications GYNOCANESTEN ® Clotrimazole

GYNOCANESTEN ® is indicated in the treatment of gynecological symptoms associated with Candida vulvovaginal infections characterized by burning, leucorrhoea, edema of the vaginal mucosa and a burning sensation during the minting act.

Mechanism of action GYNOCANESTEN ® Clotrimazole

GYNOCANESTEN ® is a drug particularly used in the clinical field to cope with the gynecological symptoms characteristic of Candida infection.

More precisely, Clotrimazole, the active ingredient of GYNOCANESTEN ®, used vaginally, maintains very high concentrations in the center, carrying out its antimicrobial activity through the inhibition of ergosterol synthesis, a key element in the structuring of the plasma membranes of the various fungi such as Candida.

More precisely, once penetrated into the pathogenic microorganism, Clotrimazole inhibits the enzyme 14 alpha-sterol dimethylase, compromising on one hand the synthesis of ergosterol and determining on the other the accumulation of methylsterol metabolites capable of interfering with the normal biological activities of membrane enzymes thus inducing the halt of the biological activities of the fungus.

Everything is also facilitated by the excellent pharmacokinetic properties that prevent the active ingredient taken via vaginal way by GYNOCANESTEN ® from reaching clinically relevant blood concentrations that are potentially responsible for drug interactions rather than side effects, such as to compromise the patient's state of health.

Studies carried out and clinical efficacy

THE CLOTRIMAZOLO AS AN ANTICANCER

Zhonghua Kou Qiang Yi Xue Za Zhi. 2010 Sep; 45 (9): 545-8.

Yet another study that demonstrates the potential antitumor effects of Clotrimazole through the inhibition of tumor cell growth of an oral squamous cell carcinoma, most likely related to inhibitory activity against the cicline-cdk complex

CLOTRIMAZOLO vs FLUCONAZOLO

J Infect Public Health. 2011 Sep; 4 (4): 195-9.

Clinical trial demonstrating that oral administration of Fluconazole may have the same efficacy as intra-vaginal Clotrimazole in the treatment of vulvovaginal candidiasis, but with an increased risk of side effects.

BIOLOGICAL MECHANISM OF THE CLOTRIMAZOLE

Antimicrob Agents Chemother. 2011 Sep; 55 (9): 4436-9.

Study of molecules that demonstrates the biological activity of antifungal agents such as Clotrimazole against Candida albicans, limiting its adhesive, invasive and damage to the vaginal epithelium.

Method of use and dosage

GYNOCANESTEN ®

Clotrimazole 2% vaginal cream;

100 mg Clotrimazole vaginal tablets.

Warnings GYNOCANESTEN ® Clotrimazole

Despite the generally safe therapy with GYNOCANESTEN ® it would be advisable in any case to consult your doctor first in order to characterize the bacterial infection and opt for the best therapeutic scheme.

The vaginal cream form, contains cetostearyl aclool, potentially responsible for contact dermatitis in atopic patients.

PREGNANCY AND BREASTFEEDING

The use of GYNOCANESTEN ® during pregnancy and in the subsequent period of breastfeeding is generally contraindicated due to the absence of clinical trials able to fully characterize the safety profile of the drug for fetal health.

Interactions

Despite the systemic absorption of Clotrimazole associated with vaginal use of GYNOCANESTEN ® is generally limited in time, it would be advisable to consider the possible interactions with tacrolimus, such as to determine an increase in the plasma concentrations of the drug with consequent increased risk of side effects.

Contraindications GYNOCANESTEN ® Clotrimazole

GYNOCANESTEN ® is contraindicated in patients who are hypersensitive to the active substance or to one of its excipients.

Undesirable effects - Side effects
