Melatonin : it is naturally synthesized in the body, in a gland placed at a diencephalic level and defined as an epiphysis or pineal gland.

The epiphysis plays a mediating role between the external environment and the endocrine activity, regulating its activity based on the photoperiod. In fact, the presence of photocopiers seems to significantly modulate the production of melatonin, inhibiting it in the presence of light and promoting it in the absence of light stimuli. All this translates into a rhythmic production of melatonin, which is maximum during the night and then significantly decreases during the day. It is therefore possible to consider this substance as the biological clock of the human organism, because by activating different types of receptors, disseminated at the encephalic level (above all hypothalamic), it can act as a sedative and hypnotic inducing sleep. The production of melatonin is maximum during the first 7 years of life, and then gradually decreases (in reality the synthesis levels remain constant, while the blood volume increases), and become minimal in the elderly, where the reduced innervation on one side and epiphyseal calcification on the other, seriously compromise the endocrine function of this gland. The oral integration of melatonin, fortunately, has been shown to temporarily reduce the endogenous secretion of this hormone, delaying the exhaustion and aging of the pineal gland and preserving its functionality.

Although there are many studies in the literature, it is still not possible to draw a complete profile of all the potential functionalities of this molecule; therefore we will describe the main biological aspects in which it is involved:

  1. Sleep / wake rhythm: by interacting with receptors disseminated at the hypothalamic level, melatonin can exert a hypnotic and sedative effect;
  2. Biological protection: exercised not only on the pineal gland, but generalized to the whole organism (given the protective action described with respect to nucleic acids subjected to chemical stress);
  3. Antioxidant processes: melatonin is directly involved in the detoxification system by oxygen and nitrogen free radicals, contributing - together with its metabolites - to total antioxidant capacities;
  4. Immune system: it is able to strengthen the body's immune defenses, antagonizing the immuno-depressive effects induced by acute stress and improving both the pro-inflammatory cytokine secretion and the differentiation and activation of the lymphocyte cell component;
  5. Metabolic pathways: recent evidence is demonstrating an important role of melatonin, and its MT1 receptor, in carbohydrate metabolism. Indeed, receptor activation appears to improve insulin sensitivity, reducing the risk of type II diabetes and associated diseases such as obesity.

Melatonin in sports

Although it is difficult to justify and describe the advantages deriving from the use of melatonin in sports, the scientific literature provides some interesting points of reflection on possible applications of this supplement:

  • Improvement of hormonal function: the role of a biological clock for melatonin could be particularly important to improve hormone secretion in individuals subjected to intense training. Although the literature is not very rich in such studies, there are some scientific evidences that correlate the increase in nocturnal GH secretion with the administration of melatonin.
  • Protective action: the antioxidant action of melatonin could be important in counteracting the oxidative damage induced by intense physical exercise.
  • Metabolic action: despite the lack of scientific evidence on men, in animal studies melatonin induced a reduction in lactate synthesis, probably associated with the previously described metabolic effect on glucose tolerance.
  • Thermoregulatory action: some evidence suggests that the drop in body temperature, recorded following the intake of melatonin, may be beneficial for athletes subjected to training and performance at high temperatures.

Although the ideas for application in the sports field are interesting, it is important to remember that the main therapeutic indication of melatonin is related to the treatment of various sleep disorders; however, there is no lack of studies that prove its efficacy in neurodegenerative, endocrine and metabolic diseases. In sport, however, there is no improvement in melatonin-dependent performance or body composition, although there is evidence of its hormone-regulating and antioxidant utility.

In most of the studies dosages ranging from 300 mcg to 5/6 mg are administered. In sports, the recommended dosage is 3 milligrams, although studies exist to support the efficacy of only 0.5 mg.
