Indications AMOX ® Amoxicillin

AMOX ® is a broad-spectrum antibiotic used clinically in antibiotic therapy for infections of organs and systems supported by Gram positive and non-penicillinase producing bacteria.

In association with pantoprazole and clarithromycin the amoxicillin can also be used in therapy for the eradication of Helicobacter Pylori.

Mechanism of action AMOX ® Amoxicillin

AMOX ®, a drug based on amoxicillin, is an antibiotic particularly used in clinical settings due to the great versatility of action that makes it effective against infections sustained by both Gram positive and negative bacteria.

The particular chemical structure, characterized by the addition of a hydroxyl group with respect to ampicillin, significantly influences the pharmacokinetic properties, giving the active principle a natural resistance to acid environments which guarantees an effective and rapid absorption through the intestine even when taken for os.

However, the microbicidal mechanism of action remains the same as that commonly described for penicillins, accomplished by inhibiting the transpeptidation reaction which prevents the formation of cross-linkages at the level of the peptidoglycan, thus reducing the stability of the bacterial wall and thus facilitating its lysis for osmotic shock.

After having exercised its bactericidal action at the level of body fluids and body tissues, amoxicillin is eliminated as it is through the kidneys .

Studies carried out and clinical efficacy

1.THE THERAPEUTIC COMPLIANCE AT THE AMOXICILLIN

Respirology. 2012 May; 17 (4): 687-92.

An interesting study that demonstrates how the possibility of marketing more stable solutions based on amoxicillin, which can reduce the daily intake frequency, can determine a significant improvement in the compliance of the therapy, increasing its effectiveness.

2 . HIGH AMOXYCILLIN DOSES IN TRIPLE THERAPY IMPROVE ERADICATION RATE

Aliment Pharmacol Ther. 2012 May 17.

Work conducted on over 100 patients suffering from gastro-enteric disorders associated with Helicobacter Pylori, which has shown that the increase in conventional amoxicillin doses can increase the efficacy of triple therapy, guaranteeing a greater degree of eradication of the microorganism.

3. HEPATIC DAY FROM AMOXICILLINA

Korean J Hepatol. 2011 Sep; 17 (3): 229-32.

Case report demonstrating the appearance of hepatocellular damage and hepatic cholestasis following the intake of amoxicillin. These cases emphasize the importance of medical supervision throughout antibiotic treatment.

Method of use and dosage

AMOX ®

Tablets of 1 g of amoxicillin;

The dosing schedule envisaged for the antibiotic treatment based on AMOX ® should be defined by your doctor based on the physio-pathological characteristics of the patient and his / her clinical picture.

The therapeutic range, generally included in adults between 1 and 3 grams of amoxicillin daily, could be reviewed in patients suffering from liver and kidney diseases or the elderly.

The use of AMOX ® must necessarily be supervised by your doctor.

Warnings AMOX ® Amoxicillin

Before taking AMOX ® it is necessary to consult your doctor and assess the possible presence of penicillin and cephalosporin allergies, whose presence would seriously compromise the patient's state of health following the intake of amoxicillin.

The doctor should also evaluate the susceptibility of the pathogen to amipicillin by antibiogram, administering the drug only in positive cases.

It is in fact useful to reiterate how the inadequate use of the drug could contribute to the establishment of drug-resistant microbial strains, therefore not sensitive to amoxicillin therapy.

The doctor should periodically check the status of liver and kidney function of the patient undergoing therapy, suspending treatment following the appearance of undesirable side effects.

PREGNANCY AND BREASTFEEDING

The use of AMOX ® in pregnancy should only be carried out in cases of real need and always under strict medical supervision.

Given the ability of amoxicillin to concentrate in breast milk, it would be advisable to avoid taking AMOX ® during breastfeeding.

Interactions

In order to reduce as much as possible the side effects of antibiotic therapy with AMOX ®, while at the same time safeguarding its therapeutic efficacy, particular attention should be paid to the simultaneous intake of:

  • Allopurinol, given the increased risk of developing allergic reactions;
  • Probenecid, responsible for the increase in blood concentrations and the hepatic and renal side effects of amoxicillin;
  • Warfarin, due to the increased risk of bleeding;
  • Methotrexate, given the increase in blood concentrations of this active ingredient induced by amoxicillin, responsible for an increase in the toxicity of the drug;
  • Ethanol and amiloride able to alter its intestinal absorption profile.

Contraindications AMOX ® Amoxicillin

The use of AMOX ® is contraindicated in patients hypersensitive to penicillins and cephalosporins or their excipients and in the course of infectious mononucleosis.

Undesirable effects - Side effects

Although the intake of AMOX ® is generally well tolerated, the allergic power of amoxicillin could determine, especially in predisposed patients, the appearance of skin eruptions, angioedema, bronchospasm, laryngospasm, hypotension and in more severe cases anaphylactic shock.

To the above mentioned side effects one could add vomit, diarrhea, anorexia, pseudo-membranous colitis, epigastric pain, hypertransaminasemia, hyperbilirubinemia, headache and dizziness, fortunately rare.

Note

AMOX ® is a prescription-only drug.
