Melatonin: also known as N acetyl 5 methoxytripamine, it is produced at the level of the epiphysis starting from the amino acid tryptophan. Its synthesis is not solely regulated by the availability of the substrate, but is affected by the activation of a key enzyme known as serotonin N acetyl transferase. The control of this enzyme is mediated by a series of biological phenomena, not yet fully known, which see the association between external stimulus and biological function. In fact, the presence in the pineal gland of cells similar to retinal photocatalysts makes it particularly sensitive to light, so much so that it can modulate its activity based on the light / dark cycle. The electromagnetic wave (light), striking these photoceptors, induces functional changes of the cell, which lead to the increase of the cyclic AMP with consequent inactivation of the enzyme Serotonin N-Acetyl Transferase, and reduction of melatonin synthesis. It can therefore be easily deduced, as the production and the secretion of melatonin is maximum in the night hours and is zeroed instead during the day. The maintenance of this rhythm and cicardiano guarantees the correct functioning of the organism and in particular of the clock that regulates sleep and wake.

The sedative and sleep-inducing effect is guaranteed by the presence of receptors for melatonin, present at the hypothalamic level and capable, once activated, of inducing the hypnotic and sedating effect that characterizes this molecule.

However, recent evidence emphasizes that melatonin is particularly active also in other parts of the body, mediating different biological actions, such as:

  1. Antioxidant effect: it has been shown as melatonin in the first place, but also the subsequent 4 metabolites that derive from its oxidation, can protect the organism from the oxidative insult induced by reactive oxygen and nitrogen species. In this case, however, the absence of enzymes capable of reducing these species leads to a net loss of the melatonin mass involved in the red-ox process.
  2. Immunostimulatory effect: melatonin has proven to be able to decisively improve the antibody response and the cytotoxic response, also enhancing the secretion of pro-inflammatory cytokines. This action could be dangerous in case of patients suffering from autoimmune or allergic diseases.
  3. Protective effect: several studies support the protective action against mitochondrial and nuclear DNA by mutagenic and potentially carcinogenic agents. Initial evidence also suggests an anti-neoplastic action, observed however exclusively in vitro and on animal models.
  4. Metabolic effect: also in this case, some studies conducted in vitro and on animal models have suggested an important metabolic action, which appears to be associated with the MT1 receptor and to be carried out by improving insulin sensitivity, especially in obese, diabetic and high fat diets.

In sports, melatonin has few fields of application, given the presence of other more effective and targeted supplements; however it is used by some athletes in an attempt to improve hormone secretion, protect the body from overtraining syndrome and strengthen the pull of antioxidant defenses.

The studies, however, do not seem very consistent and in the sporting field there is no experimentation that would allow us to draw conclusions; despite this there are works in which, following the administration of melatonin, there is an improvement in nocturnal GH secretion, an improvement in antioxidant capacity and a protection of the immune system. It must be stressed, however, that these advantages would be expressed through the reproduction of the normal circadian rhythm, avoiding subjecting the body to further stress and hormonal deregulation.

Take 4 drops of liquid equal to 1 mg of melatonin, using the dropper contained in the pack, with water or juice, in the evening 20 minutes before bedtime.

The various studies, which for several years have recommended minimum dosages of 3 mg per day, have been accompanied by new works that show the same results even for much lower dosages, even equal to 0.3 mg. The advantage of the liquid form compared to other supplements in capsules or tablets, does not materialize with the improvement of the kinetic properties, which remain unchanged, but in the simplest assumption and above all in a better modulation of the dosage, considering that on average per drop there would be about 0.25 mg of product.

Also in this case, the supplement should be taken before going to bed, so as to preserve the normal rhythm of endogenous secretion, reducing the "working" weight of the epiphysis.

Synergy - Liquid Melatonin - Natrol

Several studies seem to agree on the greater efficacy of synergy when taken together with zinc. A generalized improvement is obtained both on the strengthening of the immune defenses, on the antioxidant effect and on the metabolic aspect, improving insulin sensitivity.

Some studies show that with prolonged intake of zinc and melatonin, magnesium supplementation also becomes necessary.

The validity of the simultaneous administration of melatonin and vitamin B6, is still under observation, even if the first works show an improvement in the endogenous synthesis of this hormone, with an enhancement of the biological effects related above all to the modulation of the immune and hormonal activity .

Although recent studies have confirmed the high tolerability and safety of the active ingredient, there are cases in literature in which, even at a dose of 3 mg / day, nausea, irritability, nightmares and vascular changes have been observed.

Furthermore, acting as a mild hypnotic and sedative, melatonin may interfere with concentration. By enhancing the immune response, the substance could also worsen the condition of patients suffering from allergic or autoimmune diseases.

The product is contraindicated in cases of renal or hepatic disease, cardiovascular disease and / or hypertension, allergies and autoimmune diseases, during pregnancy, during breastfeeding, under 12 years and for adolescents not yet trained.
