Indications MACROZIT ® Azithromycin

MACROZIT ® is an antibiotic used in a medical clinic to cope with infections caused by microorganisms sensitive to macrolides and more precisely to azithromycin, often the cause of infections affecting the upper and lower respiratory tract, skin and soft tissues as well as genital apparatus.

Mechanism of action MACROZIT ® Azithromycin

MACROZIT ®, a drug based on Azithromycin, is an antibiotic belonging to the family of macrolides with 15 carbon atoms, generally used clinically in the treatment of infections sustained by Gram-negative aerobic organisms.

The therapeutic power, carried out by the inhibition of protein synthesis mediated by the interaction with the 50S ribosomal subunit responsible together with the peptidyltransferase enzyme of the lengthening of the pepetidic chain, takes place a few minutes after the oral Azithromycin intake to optimize itself after a few hours .

In this way, only after a few days of treatment, the bacteriostatic action of this macrolide is sufficient to limit the colonization and spread of the pathogen, adequately controlling the clinical symptoms.

After its activity, sometimes hampered by the onset of resistance mechanisms such as the expression of efflux pumps, the variation of the enzyme target and the enzymatic inactivation of the active principle, Azithromycin is partly eliminated also through the biliary route .

Studies carried out and clinical efficacy

1 AZITROMYCIN IN DENTISTRY

J Periodontal Res. 2012 Apr; 47 (2): 137-48.

Study demonstrating the triple role of azithromycin in periodontal pathologies: it suppresses pathogens, carries out an anti-inflammatory activity, optimizes the persistence of macrophages and fibroblasts in the affected tissue.

2. THE GASTRIC BYPASS SENSITIVELY REDUCES AZITROMYCIN ABSORPTION

J Antimicrob Chemother. 2012 Sep; 67 (9): 2203-6.

Work that demonstrates how gastric bypass can reduce the absorption of Azithromycin by reducing the area under the curve, linked to the absorption of the drug over time, by a good 1/3.

3.AZITHROMYCIN AND CARDIOVASCULAR RISK

N Engl J Med. 2012 May 17; 366 (20): 1881-90.

Very prestigious study that shows how the 5-day treatment with Azithromycin can modestly increase the number of deaths associated with cardiovascular diseases, only in the high-risk sample.

Method of use and dosage

MACROZIT ®

500 mg film-coated tablets of Azithromycin;

Generally taking 500 mg of Azithromycin once daily for three days is sufficient to control most infections caused by microorganisms sensitive to this antibiotic.

In some cases, however, the use of double dosages may be necessary.

Warnings MACROZIT ® Azithromycin

The use of MACROZIT ® should be preceded by a careful medical examination useful to verify the clinical picture of the patient, his physio-pathological conditions, the possible presence of diseases incompatible with the intake of macrolides, and more generally the appropriate prescription.

The ability of Azithromycin to lengthen the QT interval and to exacerbate the symptoms present during liver pathologies represents an important limit to the use of this antibiotic in patients suffering from liver disease, from cardiovascular diseases or possibly in therapy with cardioactive drugs.

The prolonged use of MACROZIT ® could at the same time determine the onset of intestinal pathologies and in more serious cases the pseudomembranous colitis from Clostridium Difficile, due to the alterations induced by the intestinal microflora.

MACROZIT ® contains lactose and glucose; it is therefore contraindicated in patients with lactase enzyme deficiency, lactose intolerance and glucose-galactose malabsorption syndrome.

PREGNANCY AND BREASTFEEDING

The use of Azithromycin should be limited to cases of inevitable necessity in pregnant women and in the subsequent stage of lactation.

This limitation would be determined by the ability of the active ingredient to cross the placental barrier and to accumulate in breast milk.

Interactions

In order to minimize the incidence of side effects or unexpected changes in therapeutic efficacy it would be advisable to avoid the simultaneous intake of Azithromycin and oral anticoagulants, cyclosporine, digoxin and cardioactive active ingredients.

If it is inevitable, it is always advisable to consult your doctor.

Contraindications MACROZIT ® Azithromycin

Given the potential side effects of MACROZIT ® therapy, the use of Azithromycin is contraindicated in patients suffering from hepatic, cardiac and hypersensitive patients with macrolides and related excipients.

Undesirable effects - Side effects

Although the use of MACROZIT ® is generally safe and well tolerated, in some cases symptoms such as diarrhea, nausea, vomiting, abdominal cramps, headache, irritability, sensory changes, hypotension, changes in liver function and osteo-articular pain have occurred. .

Note

MACROZIT ® is a prescription-only drug.
