L-Carnosine - Dipeptide synthesized in the liver from two amino acids, L-histidine and Beta-alanine, and particularly concentrated in skeletal muscle and in the human brain. It can also be introduced through the diet, through protein foods, to cross the gastrointestinal tract and be absorbed still intact through specific transporters (PEPT); purposely conveyed, carnosine reaches the various tissues where it can be used or hydrolysed by specific enzymes, known as carnosinases, mainly expressed in the blood, kidney and liver. Although its biological mechanism has not yet been fully characterized, carnosine has proved particularly effective in:

  1. Counteract the oxidizing activity of ROS;
  2. Dab up tissue acidity;
  3. Modulate the immune and inflammatory response;
  4. Counteract the formation of advanced glycosylation products;
  5. Protect the nervous system from aging and oxidative insult;
  6. Support visual features.

For these properties it is currently tested in the treatment of numerous diseases, from neurodegenerative to cardiovascular, from metabolic to oxidative.

The use of carnosine in sports is due to some of the aforementioned functions, and in particular to the antioxidant and buffering action:

Antioxidant action: particularly important to counteract oxidative damage induced by intense physical exercise. This results in an improvement in athletic performance and in a greater capacity for post-training recovery.

Buffering action: it becomes necessary above all during intense physical exercises, where the low concentration of oxygen induces an increase in lactic fermentation, with the production of H + and lactic acid, which is potentially harmful for its action contrasting the contractile capacities and for the possibility to increase the feeling of fatigue.

The role of carnosine in sports seems to be justified also by other properties, such as those to favor peripheral vasodilation and to increase the levels of 2, 3 diphosphoglycerate, definitely improving aerobic capacity.

Furthermore, it should be noted that the carnosine pull in athletes subjected to strength training, such as bodybuilders, is twice that of healthy untrained subjects, further emphasizing a key role in muscle functioning.

Several studies show the various benefits that can be derived from carnosine supplementation in sports, especially related to improving athletic performance. The acute administration of carnosine before training can in fact be essential for the haematic buffering action, with the consequent reduction of the sensation of fatigue. But the best results are obtained by increasing the muscle carnosine pull, with a consequent increase in maximum strength levels, medium strength levels and aerobic capacity. For this supplement, in fact, there are the same benefits recorded for beta alanine, given that both have the aim of increasing the muscular levels of this dipeptide.

In sports practice very different dosages are used ranging from 500 mg to more grams per day.

However, the buffering effect seems to be realized already at the quantities provided in a healthy diet (50 / 50mg). Also in this case it seems advisable to split the daily quota in small doses, in order to reduce the hydrolytic action induced by carnosinases and at the same time also reduce renal excretion.

With this product the 500mg a day could be satisfied with 2 simple intakes, one in the morning and one 60 minutes before training, since various studies have shown how the plasma concentrations of carnosine increase 15 minutes after ingestion, with a peak which is obtained at about 2.5 hours.

As described for beta alanine supplements, synergy with creatine has been shown to be particularly useful both in improving ergogenic and anabolic properties, recording a significant increase in muscle mass in various studies. There does not appear to be any toxic effects associated with carnosine, especially when it is consumed at doses comparable to those that would be obtained with a healthy diet.

Doses higher than 500mg, used in antitumor experiments, have proved equally safe.

In experimental models even at doses of 50mg / kg no side effects were recorded.
