Directions LOCALYN ® Fluocinolone + Neomycin

LOCALYN ® is indicated electively in the treatment of external otitis, dermo-epidermis and eczema of the ear canal.

Mechanism of action LOCALYN ® Fluocinolone + Neomycin

LOCALYN ® owes its documented efficacy to the combination of two different active ingredients with complementary therapeutic properties.

In particular:

  • Neomycin is a broad-spectrum antibiotic, belonging to the category of aminoglycosides, very active against Gram-negative bacteria, easily penetrating the bacterial membrane and inhibiting the ribosomal subunit and the resulting protein synthesis;
  • Fluocinolone on the other hand is a corticosteroid with an important anti-inflammatory activity performed on the most superficial layers of the epidermis by blocking the molecular events involved in the synthesis of inflammatory mediators such as prostaglandins, prostacyclines and thromboxanes.

The aforementioned therapeutic activities are assisted by advantageous pharmacokinetic properties which allow to significantly limit all the potential side effects linked to the systemic absorption of the two active principles.

Method of use and dosage

LOCALYN ®

Ear drops containing 0.025 g of Fluocinolone acetonide and 325000 IU of Neomicin sulfate per 100 ml of product.

Always according to medical indications, the application in the adult is recommended from 3 to 5 drops 3-4 times a day directly into the ear canal, while in children 2 drops 3 times a day.

Warnings LOCALYN ® Fluocinolone + Neomycin

The use of LOCALYN ® must necessarily be preceded by an appropriate medical consultation useful to check the prescriptive appropriateness and the possible presence of contraindications to use.

Despite the good tolerability of the product, it should be remembered that the long-term and inappropriate use of the drug could significantly increase the risk of clinically relevant adverse reactions.

This risk becomes even higher in pediatric patients for whom even the minimal systemic absorption of Fluocinolone may transiently compromise the functionality of the hypothalamic-pituitary-adrenal axis

It is recommended that you consult your doctor immediately following the appearance of undesirable effects.

PREGNANCY AND BREASTFEEDING

Given the lack of studies able to better characterize the safety profile of Flucinolone for the health of the fetus and infant, and the potential side effects of systemic corticosteroids for the unborn child, it would be advisable to avoid the use of LOCALYN ® during the pregnancy and breastfeeding.

Interactions

At the present time, known pharmacological interactions are not known, although it should be remembered that the simultaneous intake of cytochrome inhibitor drugs may increase blood concentrations of the amount of corticosteroid absorbed.

Contraindications LOCALYN ® Fluocinolone + Neomycin

The use of LOCALYN ® is contraindicated in patients who are hypersensitive to the active substance or to one of its excipients, in patients with lesions associated with viral, bacterial and fungal infections that are not adequately treated and in case of perforation of the tympanic membrane.

Undesirable effects - Side effects

The use of LOCALYN ®, above all in prolonged treatments, could determine the appearance of burning, itching, irritation, dry skin, hypertrichosis and folliculitis limited to the treated area.

Fortunately, the incidence of potential systemic side effects is rarer.
