Indications MUSCORIL ® - Tiocolchicoside

MUSCORIL ® is indicated for the treatment of acute and chronic lumbosciatica, cervico-brachial neuralgia, stubborn torticollis, post-traumatic and post-operative pain syndromes, spastic hemiparesis, Parkinson's and neurodyslectic syndrome.

Mechanism of action MUSCORIL ® - Tiocolchicoside

MUSCORIL ® is a medicinal product based on Thiocolchicoside, an active ingredient derived from Colchicoside, and has a marked muscle relaxant activity.

This activity, concentrated at the level of the central nervous system, is carried out through the binding of the active principle and its active metabolites to the GABAergic and Glycineergic receptors, on which it exercises a generally antagonizing activity, thus highlighting its complex supraspinal action in regulating muscle tone.

Both oral and parenteral administration guarantee good absorption and good biodistribution of the active ingredient which acts effectively at the central level, to be excreted, after an average half-life of 7 hours and after hepatic metabolism, mainly through the faeces.

Work that demonstrates how the association between Aceclofenac, Paracetamol and Thiocolchicoside can determine a significant reduction in low back pain, compared to monotherapies, resulting among other things very well tolerated.

IPERSENSIBILITY 'IGE MEDIUM AT TIOCOLCHICOSIDE

Case report that reveals the appearance of hypersensitivity to thiocolchicoside, observing IgE-mediated reactions also of severe intensity, so as to determine anaphylaxis to the aforementioned active principle.

Method of use and dosage

MUSCORIL ®

Hard capsules of 4 - 8 mg of Thiocolchicoside;

Tiocolchicoside 9 mg tablets;

Injectable solution for intramuscular use from 4 mg of TIocolchicoside to 2 ml of solution.

The definition of the dosage of the relative therapeutic scheme is up to the doctor after having carefully evaluated the patient's overall health and the severity of his clinical picture.

It is generally recommended to take 2 capsules of 4 mg twice a day or one of 8 mg twice a day or 2 vials for intramuscular use a day, 12 hours apart.

The use of this drug is contraindicated in children under 15 years of age.

Warnings MUSCORIL ® - Tiocolchicoside

Therapy with MUSCORIL ® must necessarily be preceded by a careful medical examination in order to assess the prescriptive appropriateness and the possible presence of contraindications to the use of the drug.

Particular caution in the administration of Thiocolchicoside must be guaranteed in patients suffering from epilepsy, due to the increased risk of convulsions, and in patients suffering from gastroenteric diseases such as diarrhea.

The entire therapeutic procedure should be supervised by medical personnel, given the increased risk of episodes of vaso-vagal syncope, following intramuscular administration.

MUSCORIL ® in capsules contains lactose among its excipients, therefore it is not indicated in patients with lactase enzyme deficiency, glucose / galactose malabsorption syndrome and galactosemia.

MUSCORIL ® in orodispersible tablets instead presents among its excipients aspartame, which as a source of phenylalanine, could be harmful in patients suffering from phenylketonuria.

PREGNANCY AND BREASTFEEDING

Although there are no clinical trials able to better characterize the safety profile of Thiocolchicoside for the health of the fetus and infant, some experimental evidences that show a toxic reproductive potential linked to the active principle, suggest the extension of the aforementioned contraindications to the use of MUSCORIL ® also to pregnancy and subsequent breastfeeding period.

Contraindications MUSCORIL ® - Tiocolchicoside

The use of MUSCORIL ® is contraindicated in patients suffering from flaccid paralysis, muscular hypotonia, in patients who are hypersensitive to the active substance or to one of its excipients and during pregnancy and lactation.

Undesirable effects - Side effects

The use of MUSCORIL ® is generally safe and well tolerated, although sometimes associated with the risk of gastrointestinal adverse reactions such as nausea, vomiting and diarrhea, hypersensitivity reactions and drowsiness.
