Indications MAXAQUIN ® Lomefloxacin

MAXAQUIN ® is an antibiotic used clinically in the treatment of urinary tract infections and lower respiratory tract supported by microorganisms sensitive to Fluoroquinolones and in particular to Lomefloxacin.

Mechanism of action MAXAQUIN ® Lomefloxacin

Lomefloxacin is a semi-synthetic antibiotic belonging to the second-generation Fluoroquinolones category, active both against Gram-negative and Gram-positive bacteria, and characterized by the marked bacteriostatic activity exerted by the inhibitory capacity against some bacterial enzymes, such as DNA gyrase and DNA topoisomerases, normally involved in the replication of bacterial DNA.

The bacteriostatic activity, effective even at minimum concentrations, also extends to the lower urinary tract thanks to some pharmacokinetic characteristics that allow the active principle to reach unchanged the urinary environment

From the pharmacokinetic point of view in fact Lomefloxacin, taken orally, is rapidly absorbed at the intestinal level reaching the maximum plasma concentration in just 90 minutes.

Distributed between the various tissues, with the exception of the nervous system, given the inability to cross the blood-brain barrier with intact meninges, it performs its therapeutic action, being minimally metabolised by liver enzymes and therefore being excreted unchanged through the urine .

These characteristics make the administration of Lomefloxacin effective and highly reproducible with the exception of patients suffering from renal function deficiency, for which unfortunately continuous dosage adjustments are required.

Study that compares the therapeutic efficacy of Lomefloxacin to that of Ciprofloxacin in the treatment of chronic bacterial prostatitis, showing similar results both in terms of collateral and therapeutic effects

LOMEFLOXACINE AND BACTERIA DIVIDE

Work demonstrating the effectiveness of the use of Lomefloxacin topically in reducing clinical complications associated with bacterial conjunctivitis, thus demonstrating antibiotic efficacy even when used topically.

DIFFUSION OF MULTIRESISTIC STRIPS

Interesting study that demonstrates for the first time the onset of mechanisms of resistance to fluoroquinolones and in particular to Lomefloxacin in Chlamydia Trachomatis, thus describing multiresistant strains.

Method of use and dosage

MAXAQUIN ®

Lomefloxacin 400 mg tablets.

Generally the suggested dosage is one tablet per day for 7-14 days depending on the type of infection and its clinical severity, taking care to continue therapy for at least 2-3 days despite symptom remission.

The therapeutic scheme, in any case, should also be set by the competent doctor based on the physiopathological conditions of the patient.

Warnings MAXAQUIN ® Lomefloxacin

In order to minimize the occurrence of clinically relevant side effects, thereby safeguarding the efficacy and safety of the therapy, it is recommended to:

  • Avoid exposure to ultraviolet radiation, given the photosensitivity induced by Lomefloxacin;
  • Request medical consultation and supervision when suffering from kidney disease;
  • Use MAXAQUIN ® with caution in case of enzyme deficiency of glucose 6 phosphate dehydrogenase.

PREGNANCY AND BREASTFEEDING

Given the pharmacokinetic characteristics of Lomefloxacin and given the absence of studies able to evaluate the safety profile of the drug on the exposed fetus, the use of MAXAQUIN ® during pregnancy and in the subsequent period of breastfeeding is not recommended.

Interactions

In light of the various pharmacokinetic studies present in the literature, it would be appropriate for the patient receiving MAXAQUIN ® to avoid the simultaneous use of:

  • Foods, supplements and drugs containing divalent metals, given the ability to form chelating complexes, responsible for reducing the therapeutic efficacy of the antibiotic;
  • Tiazanidine, methotrexate, theophylline, xanthine and phenytoin due to the altered tubular secretion induced by Fluoroquinolone;
  • Oral anticoagulants, due to the increased anticoagulant activity induced by Lomefloxacin.

Contraindications MAXAQUIN ® Lomefloxacin

The use of MAXAQUIN ® is contraindicated in patients who are hypersensitive to the active ingredient or to one of its excipients, during pregnancy and in the subsequent period of breastfeeding.

Undesirable effects - Side effects

The use of MAXAQUIN ®, especially when prolonged over time, could cause the appearance of nausea, vomiting, diarrhea and in severe cases pseudomembranous colitis, headache, irritability, asthenia and sleep disorders, photosensitivity, renal and hepatic impairment and osteo-articular disorders.
